PTP-PEST Antibody (AG25) is a mouse monoclonal IgG1 antibody that detects PTP-PEST in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). PTP-PEST, a 780 amino acid cytosolic nonreceptor protein encoded by a gene located on chromosome 7q11.23, plays a critical role in regulating cellular signaling pathways by modulating tyrosine phosphorylation. PTP-PEST is predominantly expressed in various hemopoietic cell types, including B cells and T cells, where PTP-PEST is involved in essential processes such as cellular growth and differentiation. Notably, PTP-PEST interacts with several key signaling molecules, including Shc, paxillin, Csk, and Cas, facilitating the dephosphorylation of these proteins and thereby influencing pathways like the Ras signaling cascade. PTP-PEST′s ability to regulate c-Abl activity through dephosphorylation represents a significant mechanism of action, highlighting PTP-PEST′s importance in cellular signaling. Furthermore, PTP-PEST is crucial for cytoskeletal organization, promoting the turnover of focal adhesions necessary for cell migration and regulating the proline, serine, threonine phosphatase interacting protein (PSTPIP)-mediated cleavage furrow formation during cell division.
