PTN Antibody (H-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects PTN protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-PTN antibody (H-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Pleiotrophin (PTN) is a heparin-binding growth factor that plays a crucial role in cellular processes such as cell proliferation, differentiation, and migration, making PTN significant in developmental biology and tissue repair. PTN protein is synthesized as a 168 amino acid precursor that is cleaved to yield a 136 amino acid active form, exhibiting approximately 50% identity with midkine (MK) and maintaining conservation of all ten cysteine residues, which are essential for structural integrity and function. PTN is predominantly expressed in various cell types, including osteoblasts, chondrocytes, and neurons, and is particularly important during fetal development, where PTN supports the growth and differentiation of neural tissues. In adult organisms, PTN expression is largely restricted to the brain, highlighting PTN′s specialized role in neurodevelopment and potential implications in neurodegenerative diseases.

Hi I am using this antibody for western blot in NT2 cells and I detect two bands around molecular weight 25KDa and 17KDa. I also noticed you have shown the western blot image of PTN in NT2 cells. Could you help us verify which is the correct PTN band.

Thank you for your question. PTN has a predicted molecular weight of ~19 kDa, however we typically observe PTN in cell lines between 18-25 kDa. The Western blot data published on our website is representative of what we observe using the antibody in cell lines tested. We recommend you include positive and negative controls in your evaluation of the antibody in your samples to evaluate the specificity of your results. Please contact our Technical Service Department for support in troubleshooting the antibody.

I am using PTN (H-6): sc-74443 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
