PSD-95 Antibody (7E3) is a mouse monoclonal IgG1 kappa light chain antibody that detects PSD-95 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Anti-PSD-95 antibody (7E3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PSD-95, also known as SAP 90, plays a crucial role in organizing synaptic signaling complexes and is essential for proper functioning of excitatory synapses. PSD-95 is primarily located at postsynaptic density of neurons, where PSD-95 interacts with various receptors and scaffolding proteins, facilitating clustering of glutamate receptors and other signaling molecules. This localization is vital for synaptic plasticity, a process that underlies learning and memory. PSD-95 forms complexes with other proteins, such as NMDA and AMPA receptors, underscoring PSD-95′s importance in modulating synaptic strength and transmission. Additionally, PSD-95′s interaction with proteins like tumor suppressor APC suggests a role in regulating cellular growth and signaling pathways, highlighting significance beyond synaptic function.

I used the rat hippocampus protein sample and saw two bands with the Ab, as the picture shows. why?

What is the dilution for WB in tissue?

Thank you for your question. PSD-95 (7E3) is recommended for detection of PSD-95 of mouse, rat and human origin by Western Blotting (starting dilution 1:200, dilution range 1:100-1:1000).

Please, which dilution for IF in cells?

Thank you for your question. For immunofluorescence applications with PSD-95 Antibody (7E3): sc-32290 we recommend a dilution range of 1:50-1:500 with a starting dilution of 1:50.

This antiboby can be used for the immunohistochemistry-paraffin embedded tissue. (IHC-P)

Thank you for your question. We have not yet tested this antibody, PSD-95 (7E3): sc-32290, on formalin-fixed paraffin-embedded tissue sections and so IHC(P) is not currently a recommended application. We do recommend this antibody for IF or immunofluorescent staining of fresh or frozen tissues or cells fixed in acetone or methanol.

Thank you for your question. PSD-95 Antibody (7E3) is a mouse monoclonal IgG1 with a kappa light chain. We recommend using one of our new Mouse IgG kappa Binding Proteins in place of a conventional secondary antibody. For example, for Western blotting we would recommend our HRP conjugated, m-IgGκ BP-HRP: sc-516102, (https://www.scbt.com/scbt/product/m-igg-kappa-bp-hrp). A complete list of Mouse IgG Binding Proteins is available on our website using the link below. https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-130b1ry Any conventional anti-mouse IgG secondary antibody would also work just fine with this primary antibody.
