PSCA Antibody (7F5) is a mouse monoclonal IgG1 kappa light chain antibody that detects PSCA protein of human origin by western blotting (WB), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). PSCA (7F5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Prostate stem cell antigen (PSCA) is a 123 amino acid glycosylated protein that belongs to the Thy-1/Ly-6 family of glycosyl-phosphatidylinositol (GPI)-anchored cell surface antigens, which play crucial roles in cell signaling and adhesion. PSCA gene is located on chromosome 8q24.3, where PSCA is predominantly expressed in prostate and placental tissues. Notably, PSCA is overexpressed in a significant number of prostate cancers, and PSCA transcripts are also found in high levels in urothelial tumors, indicating potential as a biomarker for both urothelial and gastric tissue carcinogenesis. The interaction of PSCA with specific proteins, such as c-myc, which is also located on chromosome 8q and is known for its role in cell proliferation and cancer progression, highlights PSCA′s importance in cancer biology. Among prostate cancer cell surface antigens, PSCA is expressed in over 80% of prostate carcinomas and is associated with certain phenotypes, including prostate cancer bone metastases, making PSCA a valuable target for therapeutic and diagnostic applications.

What is the pH value of antigen repair in IF test.

Thank you for your question. The PH is related to the mode of antigen repair. You can refer to the following repair methods. Heat treatment (recommended method): Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.

Does this antibody work against mouse PSCA?

Is this antibody good for WB?

Thank you for your question. PSCA Antibody (7F5): sc-80654 is in fact recommended for detection of PSCA of human origin by WB, as well as IF, IHC(P) and FCM. There is published data for the use of this antibody in western blots as well (PMID 22133690).

Does the antibody only consists of the light chain of IgG? Can it be coupled to protein G coated magnetic beads?
