PRPS1/2/3 Antibody (A-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects PRPS1/2/3 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). PRPS1/2/3 (A-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PRPS (phosphoribosyl pyrophosphate synthetase) proteins are crucial enzymes that catalyze the synthesis of phosphoribosyl pyrophosphate (PRPP), a key substrate in nucleotide biosynthesis. The three human PRPS isoforms, encoded by distinct genes, include PRPS1 and PRPS2, which are widely expressed across various tissues, while PRPS3 is primarily found in the testis. The synthesis of PRPP from MgATP and ribose-5-phosphate, requiring inorganic phosphate and magnesium as cofactors, is vital for the production of nearly all nucleotides, underscoring the importance of PRPS1 and PRPS2 in purine metabolism and overall cellular function. Mutations in the PRPS1 gene can lead to PRPS superactivity, resulting in conditions such as gout due to the overproduction of purine nucleotides, uric acid, and PRPP. Conversely, reduced PRPS1 activity due to mutations can cause severe disorders like ARTS syndrome and Charcot-Marie-Tooth disease X-linked recessive type 5 (CMTX5), highlighting the critical role of PRPS proteins in maintaining nucleotide balance and cellular health.
