PRPK Antibody (F-9) is a mouse monoclonal IgG1 antibody that detects PRPK in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PRPK, also known as TP53RK, is a 253 amino acid protein kinase that plays a crucial role in the regulation of the tumor suppressor protein p53 by phosphorylating Ser15, which enhances p53′s stability and activity. This phosphorylation is vital as p53 can effectively upregulate genes associated with growth arrest and apoptosis in response to cellular stress, thereby influencing critical processes such as programmed cell death, cell differentiation, and cell cycle control. The interaction between PRPK and p53 is significant because unphosphorylated p53 is targeted for degradation via the ubiquitin-proteasome pathway, potentially leading to unchecked cell proliferation. PRPK is predominantly localized in the nucleus and exhibits high expression levels in the testis, with lower levels found in the heart, kidney, and spleen, highlighting PRPK′s importance in cellular functions and potential implications in cancer biology. Anti-PRPK antibody (F-9) is an essential tool for researchers investigating the intricate mechanisms of p53 regulation and tumor suppression.

Can sc-100350: PRPK (F-9) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: TinTin
Thank you for your question. Yes, sc-100350: PRPK (F-9) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
