PRP6 Antibody (C-12) is a mouse monoclonal IgM antibody that detects PRP6 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PRP6 plays a crucial role in the assembly of pre-mRNA spliceosomes, specifically by facilitating the interaction between small nuclear ribonucleoproteins (snRNPs) U4/U6 and U5 to form the essential [U4/U6.U5] tri-snRNP complex. This interaction is vital for the splicing process, which is necessary for the maturation of pre-mRNA into functional mRNA, thereby influencing gene expression and cellular function. PRP6 is predominantly localized in discrete subnuclear regions, mirroring the localization patterns observed in mammalian splicing components, which underscores PRP6′s importance in the regulation of splicing dynamics. Notably, mammalian PRP6, isolated from HeLa nuclear extracts, shares conserved tetrarico peptide repeats with yeast Prp6p, establishing PRP6 as the mammalian homolog. Unlike yeast Prp6p, which is specific to U4/U6, human PRP6 interacts with both U5 and U4/U6 snRNPs through protein-protein interactions, effectively bridging these two critical components of the spliceosome. This multifaceted role of PRP6 not only highlights PRP6′s significance in RNA processing but also positions PRP6 as a key player in the intricate network of gene regulation.
