PrP Antibody (8B4) is a mouse monoclonal IgG1 kappa light chain antibody that detects the prion protein (PrP ) of mammalian origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA). PrP (8B4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The prion protein plays a crucial role in neuroprotection and cellular signaling, and misfolding is central to the pathogenesis of prion diseases, which are fatal neurodegenerative disorders. The normal cellular form of PrP (PrPc) is predominantly located in the brain, where PrPc is involved in synaptic function and neuroprotection. However, when PrPc undergoes conformational changes to form the disease-associated isoform (PrPSc), PrPSc becomes resistant to proteolytic degradation, leading to the accumulation of toxic aggregates that disrupt cellular function and trigger neurodegeneration. The presence of PrPSc in the brains of affected animals, such as those with scrapie, bovine spongiform encephalopathy, and Creutzfeldt-Jakob disease, highlights the importance of monitoring PrP levels and conformations in research and diagnostic applications. Anti-PrP antibody (8B4) is an essential tool for studying these critical processes and understanding the mechanisms underlying prion diseases.

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, PrP (8B4): sc-47729 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
