Prohibitin Antibody (E-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects Prohibitin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Prohibitin antibody (E-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Prohibitin is an evolutionarily conserved protein that plays a crucial role in regulating the cell cycle, particularly by inhibiting the G1 to S phase transition, which helps maintain proper cellular proliferation and prevent uncontrolled growth. Prohibitin primarily localizes in the inner mitochondrial membrane, where this protein contributes to mitochondrial function and acts as a tumor suppressor by interacting with retinoblastoma protein and inhibiting E2F transcription factors in response to specific signaling pathways. Prohibitin also participates in B cell maturation at the plasma membrane, demonstrating diverse roles in cellular processes. Human prohibitin genes reside on chromosome 17q21 and show expression in various tissues, with expression levels fluctuating throughout the cell cycle, suggesting complex regulatory mechanisms including both transcriptional and post-translational modifications. Mutations in Prohibitin genes have been linked to breast cancer development and progression in over 80% of analyzed cell lines, highlighting significant implications in cancer biology.

I was using the Prohibitin Antibody (H-80): sc-28259 which was discontinued and replaced for this antibody. However, using the same samples, the E-5 antibody gave me 2 bands instead of 1, which are between the 25 and the 35 kDa markers.

How can Prohibitin (E-5): sc-377037 monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Prohibitin (E-5): sc-377037 monoclonal antibody is currently available conjugated to agarose (sc-377037 AC), HRP (sc-377037 HRP), phycoerythrin (sc-377037 PE), fluorescein (sc-377037 FITC), Alexa Fluor® 488 (sc-377037 AF488) or Alexa Fluor® 647 (sc-377037 AF647).

Rated 4 out of 5 by from Several bands in WB and strong signal in IFI got three strong bands in WB (1:200 in 5% BSA) for several liver cancer cell lines, although I only expected one. I used the antibody 1:50 in IF (PFA-fixed HuH7 cells) and got a strong and convoncing signal.
