SEE ALSO...

PRL-3 Antibody (318) is a mouse monoclonal IgG1 kappa light chain antibody that detects PRL-3 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-PRL-3 antibody (318) is available in both non-conjugated and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. PRL-3, also known as Protein Tyrosine Phosphatase Type IVA, member 3 (PTP4A3), plays a crucial role in cellular signaling and is primarily expressed in heart and skeletal muscle, as well as in the epithelial cells of the small intestine, where PRL-3 associates with the cell plasma membrane. PRL-3 localization allows for the regulation of key signaling pathways that influence cell growth and differentiation. Notably, PRL-3 overexpression has been linked to the inhibition of angiotensin-II induced calcium mobilization, which is vital for various cellular functions, and PRL-3 has been implicated in promoting colorectal cancer metastasis, making PRL-3 a potential therapeutic target for cancer treatment.

just to make sure before ordering, is this a whole antibody (IgG) with a molecular weight of 150 Kda or is the size based on the kappa chain only.

Asked by: Ziyad
Thank you for your question. This is a regular complete IgG antibody with the standard ~150kDa size. The light chains for this antibody are kappa.

I got a band at approx. 40 kDa and 30 kDa but not on 20 kDa with Ab 1:200 (also 1:100) and 40 µg of DLD1 whole cell lysate. Can you please get feedback to this?

How much of recombinant PRL-3 should I use for ELISA to get a signal with sc-130355 antibody (if using as recommended 1:30 dilution) at least OD450nm ~ 1.0?

Asked by: Elas
Thank you for your question. For using this antibody for solid-phase ELISA we recommend a starting dilution of 1:30 and a dilution range of 1:30-1:300. The amount of recombinant PRL-3 that is required is highly dependent on the experimental setup of your ELISA. Please contact our technical service for assistance.

How much protein do you recommend loading on the gel for Western blot detection using PRL-3 (318): sc-130355 monoclonal antibody?

Asked by: jenniferc
We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane. A link to our complete Western blotting protocol is provided here for your reference. https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
