PRAT4B Antibody (E-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects PRAT4B protein of human origin and Cnpy4 protein of mouse and rat origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-PRAT4B antibody (E-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PRAT4B, also known as CNPY4 (canopy 4 homolog), is a 248 amino acid protein characterized by the presence of a saposin B-type domain and is a member of the canopy family. PRAT4B is primarily localized to the endoplasmic reticulum and is ubiquitously expressed, with the highest levels found in the lung, spleen, thymus, and uterus. PRAT4B plays a crucial role in regulating Toll-like receptor 4 (TLR4) expression on the cell surface; while PRAT4B does not alter total cellular levels of TLR4, PRAT4B significantly decreases surface expression, indicating involvement in modulating immune responses. PRAT4B can associate with TLR4 either alone or in conjunction with MD2, but PRAT4B does not interact with MD2 alone or with TLR2, another member of the Toll-like receptor family. PRAT4B expression is markedly elevated in patients with sepsis, particularly in severe cases, suggesting a potential role in the development of abnormal inflammatory responses in pediatric sepsis. The gene encoding PRAT4B is located on human chromosome 7q22.1, highlighting significance in both health and disease contexts.
