PPARγ Antibody (E-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects PPARγ protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PPARγ (E-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. PPARγ monoclonal antibody (E-8) is recommended for detecting both PPARγ1 and PPARγ2 gene products. PPARγ, a member of the nuclear hormone receptor superfamily, plays a crucial role in regulating gene expression related to insulin sensitivity, adipocyte differentiation, and lipid metabolism. Formation of heterodimers with retinoid X receptors (RXRs) allows modulation of gene transcription involved in critical metabolic processes, influencing pathophysiology of conditions such as obesity, diabetes, and cardiovascular diseases. PPARγ regulates inflammation and influences macrophage activity, which is vital for maintaining immune homeostasis. PPARγ mouse monoclonal antibody (E-8) supports research studying implications in metabolic disorders and interactions with various ligands, including prostanoids and thiazolidinediones, which can activate transcriptional activity. Pro12Ala polymorphism in PPARγ2 gene has been associated with altered transactivation capabilities and may impact immune response to oxidized low-density lipoprotein, linking to type 2 diabetes and abdominal obesity.

Does this product react with the human placenta?

Does HEK293 cells express this protein or could HEK293 cells be stimulated to express it?

Thank you for your question. Unfortunately we are unable to answer the expression level in HEK293 cells specifically, but this information is probably available in literature, so I would recommend looking in pubmed. We do have data in many cell lines, and we do see PPARγ expression without stimulation in most cases.

The bands we are obtaining in a WB of PAG with this antibody are around 45kDa, really close to the actin band. I am using a gel concentration of 8%. Is this band that I am obtaining PPAR gamma?? Is there any publication that can prove me it is OK?

Thank you for your question. Yes, PPARgamma has been reported at several different sizes, ranging down to 43kDa and up to 67kDa. I would suggest not using actin as a loading control when you are working with PPARgamma. GAPDH or Vinculin may be better so as to not be in the same range as your target protein.

hi, what blocking buffer that i should use with this antibody? is it better with BSA or non-fat milk?

Thank you for your question. For Western blotting with chemiluminescent detection (HRP), we recommend using 5% nonfat milk + TBS + 0.05% Tween-20 (available pre-made as, TBS Blotto A: sc-2333). For Western blotting with near-infrared (NIR) detection, we recommend using our UltraCruz® Blocking Reagent: sc-516214. Yes. We recommend using a directly conjugated mouse monoclonal primary antibody. Our PPARγ Antibody (E-8) is currently available conjugated to AF405, AF488, AF647, Biotin, FITC, HRP, PE, and TRITC.
