PNMTase Antibody (C-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects PNMTase protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-PNMTase antibody (C-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The phenylethanolamine N-methyltransferase (PNMT/PNMTase) enzyme plays a crucial role in the synthesis of epinephrine from norepinephrine, marking the final step in catecholamine biosynthesis, which is vital for the body′s response to stress and regulation of blood pressure. The human PNMT protein consists of 282 amino acids and exhibits significant homology with tyrosine hydroxylase, indicating a shared evolutionary pathway in catecholamine metabolism. PNMT expression is intricately regulated by hormonal and neural stimuli, as PNMT promoter contains sequences responsive to cholinergic and depolarization signals. Notably, cortisol and early growth response protein 1 (Egr-1) enhance PNMT expression, thereby influencing adrenaline production in pheochromocytomas, while protein kinase A up-regulates and protein kinase C down-regulates PNMT expression. Additionally, PNMT gene undergoes alternative splicing through a mechanism known as intron retention, resulting in two splice variants that are differentially expressed in a tissue-specific manner and regulated by glucocorticoids in the brain. The intron-less mRNA is predominantly expressed in the adrenal glands throughout development, while the intron-retained variant is constitutively expressed into adulthood. Importantly, variations in PNMT gene have been linked to an increased susceptibility to the sporadic form of early-onset Alzheimer′s disease, highlighting PNMT significance in both physiological and pathological contexts.
