PNAd Antibody (E-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects PNAd in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-PNAd antibody (E-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PNAd, also known as Protein N-terminal asparagine amidohydrolase, consists of 310 amino acids and is encoded by the human NTAN1 gene. PNAd plays a crucial role in the side-chain deamidation of N-terminal asparagine residues to aspartate, a process essential for the ubiquitin-dependent degradation of intracellular proteins that begin with a methionine-asparagine sequence. The conversion of N-terminal asparagine to aspartate by PNAd facilitates subsequent arginylation and polyubiquitination of these proteins, marking them for degradation according to the N-end rule, thereby regulating protein turnover and maintaining cellular homeostasis. PNAd does not act on substrates with internal or C-terminal asparagines, nor does PNAd modify glutamine residues, highlighting specificity and significance in cellular processes.
