PLZF Antibody (D-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects PLZF protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-PLZF antibody (D-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PLZF protein, also known as ZBTB16 or ZNF145, plays a crucial role in transcriptional regulation and is involved in the development of various cell types, particularly in the immune system. PLZF structure is characterized by five zinc finger motifs and a broad-complex, POZ (or BTB) domain, which are essential for function as a transcription factor. PLZF BTB/POZ domain facilitates protein-protein interactions, allowing formation of complexes with other transcriptional regulators, thereby influencing gene expression. PLZF has been implicated in the regulation of genes associated with tumor suppression, and dysregulation is linked to several cancers, including acute promyelocytic leukemia, where PLZF forms a fusion protein with retinoic acid receptor alpha (RARA), leading to altered transcriptional activity. Understanding PLZF structural features and interactions is vital for elucidating roles in cellular processes and potential as a therapeutic target in cancer treatment.

Is this antibody can be used for Flow cytometry. If yes then what is the dilution. Do you also have a small pack of this antibody.

Thank you for your question. Yes, we are aware of several publications citing use of this PLZF Antibody (D-9): sc-28319 in flow cytometry (FCM). For example, see PMID: 22473038. We recommend using 1 µg of fluorochrome-conjugated antibodies in 100 µl of the prepared cell suspension (equal to 1 million cells) to each tube. What is the recommended detection reagent for this mouse primary antibody since SCBT no longer sells traditional secondary antibodies?

Antigen retrieval 12 min in Na citrate. Permeabilization 30 min 0.2%Triton X-100/PBS. Blocking peroxidase 30 min. Primary 1:200 ON. Secondary 1:200 1h.
