Plk Antibody (3F8) is a mouse monoclonal IgG1 antibody that detects Plk in mouse, rat, and human samples through applications such as western blotting (WB) and immunoprecipitation (IP). Plk, or polo-like kinase, is a crucial serine/threonine kinase that plays a significant role in cell cycle regulation, particularly during mitosis. Plk is essential for the proper functioning of the mitotic spindle, a structure that segregates chromosomes during cell division. Plk protein accumulates during the S and G2 phases of the cell cycle, with levels and catalytic activity peaking at the onset of mitosis, which is vital for ensuring accurate chromosome segregation and preventing aneuploidy. Following mitosis, Plk levels rapidly decline, highlighting a transient yet critical role in cell division. Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
