Plectin Antibody (10F6) is a mouse monoclonal IgG1 kappa light chain antibody that detects Plectin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Plectin antibody (10F6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Plectin is a crucial cytoskeletal protein that plays a significant role in maintaining cellular integrity by crosslinking intermediate filaments to microtubules and microfilaments, thereby stabilizing the cytoplasm. Plectin′s ability to anchor intermediate filaments to both plasma and nuclear membranes is vital for cellular architecture and function. Plectin interacts with key proteins such as Lamin B and vimentin, with these interactions being modulated by various protein kinases. For instance, phosphorylation by protein kinase A or protein kinase C reduces binding affinity to Lamin B, while phosphorylation by protein kinase A enhances interaction with vimentin. Plectin serves as a substrate for p34cdc2 kinase, highlighting involvement in cell cycle regulation. Several alternative splice isoforms of Plectin exist, and mutations in the human Plectin gene are associated with epidermolysis bullosa simplex with muscular dystrophy (EBS-MD), underscoring importance in both structural and pathological contexts.
