Pleckstrin Antibody (25) is a mouse monoclonal IgG1 kappa light chain antibody that detects Pleckstrin protein of mouse, rat, and human origin by western blotting (WB) and immunoprecipitation (IP). Anti-Pleckstrin antibody (25) is available as the non-conjugated format. Pleckstrin plays a crucial role in cellular signaling, particularly in platelets and neutrophils, where Pleckstrin acts as a major substrate for protein kinase C (PKC) activation. Upon PKC activation, Pleckstrin undergoes rapid phosphorylation, which is essential for mediating inflammatory responses. Pleckstrin features a unique structure that includes two pleckstrin homology (PH) domains at N and C termini, which facilitate interactions with both proteins and lipids. This structural characteristic allows Pleckstrin to translocate to cellular membranes and interact with the cytoskeleton, thereby enhancing its role in targeting proteins or lipids to sites of action during immune responses. Pleckstrin is highly expressed in human neutrophils and is phosphorylated in response to inflammatory stimuli, likely by nonconventional PKC isoforms such as delta or zeta. The conformational changes induced by phosphorylation enable Pleckstrin to effectively contribute to the microbicidal response, underscoring its importance in immune function.

Hi, Can I use this antibody to detect Pleckstrin of mouse origin by immunofluorescence?

Thank you for your question. This antibody is currently only recommended for Western blotting of Pleckstrin in mouse, rat and human samples. We do not yet have data or citations using this antibody for immunofluorescent (IF) staining, and so it is not a recommended application at this time. Please check back for updates!

How much sample do you recommend loading on the gel for Western blot detection using Pleckstrin (25): sc-136042?
