PIP2 Antibody (PIP2 2C11) is a mouse monoclonal IgM antibody that detects PIP2 protein of mouse, rat, and human origin by immunoprecipitation (IP) and immunofluorescence (IF). Anti-PIP2 antibody (2C11) is available as a non-conjugated format. Phosphatidylinositol 4,5-bisphosphate (PIP2) is a crucial low-abundance membrane phospholipid that plays a significant role in various cellular processes, including membrane trafficking, plasma membrane-cytoskeleton linkages, and synaptic vesicle recycling. PIP2′s importance extends to second messenger signaling, cell adhesion, motility, and regulation of proteins involved in phospholipid metabolism. PIP2 stimulates Ca2+-activated ATPase, suggesting that fluctuations in cellular PIP2 concentrations may be vital for regulating calcium transport within the endoplasmic reticulum. PIP2 interacts with the taste transduction ion channel TRPM5, highlighting a role in sensory signaling pathways. Hormone-regulated PIP2 levels serve as an effector for multiple downstream proteins, indicating a central role in cellular signaling networks. PIP2 may act as a second messenger in controlling cytoskeleton-membrane adhesion, underscoring multifaceted functions in maintaining cellular integrity and communication.

Is there any recommended protocol (especially for fixation and permeabilization) to stain PIP2 using sc-53412 and sc-516140? I have no experience with IF of lipid components and wonder how it would be different from the normal IF that targets proteins.

Yes, we recommend using the azide-free form for neutralization studies, sc-53412 L. The concentration provided of the azide-free form is 200 µg in 0.1 ml.
