SEE ALSO...

PIF1 Antibody (F-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects PIF1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). PIF1 (F-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PIF1 functions as a single-stranded DNA-dependent ATPase and helicase, unwinding DNA in a 5′ to 3′ direction, which is essential for DNA replication and telomere length maintenance in organisms such as Saccharomyces cerevisiae. This function helps prevent genomic instability, which can lead to various diseases, including cancer. PIF1 is highly conserved across species, and mutations can result in telomere elongation, indicating a role as a catalytic inhibitor of both new telomere formation and elongation. In humans, PIF1 preferentially binds to telomeric DNA, where PIF1 inhibits telomerase activity, thereby playing a significant role in cellular aging and tumor suppression. PIF1 counteracts the activity of the RecQ homolog Sgs1 helicase, and PIF1 degradation is regulated by the ubiquitin-26S proteasome pathway, highlighting an importance in maintaining genomic integrity.

There are two isoforms of human PIF1 gene product, so does the antibody sc-48377 recognize both isoforms? The reference is listed below. doi: 10.1248/bpb.30.1685. PMID: 17827721.

Asked by: Robert
Thank you for your question. PIF1 Antibody (F-10): sc-48377 has been raised against full length PIF1 of mouse origin. No epitope mapping has been performed as of yet, the exact binding site on its target protein is not known, so we are not sure it can recognize both isoforms. If you have any other questions, please contact our Asian Technical Service team at (+86) 021-60936350 or asia@scbio.cn or by live chat on our website.

When should the TransCruz reagent sc-48377 X be used instead of the standard form sc-48377?

Asked by: Professor Griffin
Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
