PIBF Antibody (C-9) is a mouse monoclonal IgG1 antibody that detects Progesterone-Induced Blocking Factor 1 (PIBF) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PIBF plays a crucial role during pregnancy, as PIBF is synthesized in response to progesterone by progesterone receptor-positive T lymphocytes, predominantly gamma-delta T cells. PIBF (C-9) antibody targets a protein essential for modulating the immune response, particularly by inhibiting the activity of natural killer (NK) cells, which prevents the release of perforin and subsequent lysis of target cells. Notably, PIBF expression is significantly elevated in healthy pregnant women compared to those at risk for premature pregnancy termination, highlighting PIBF′s importance in maintaining pregnancy. The full-length PIBF protein is primarily localized in the nucleus, where PIBF is believed to function as a transcription factor, while shorter isoforms, which are secreted upon cell activation, may act as cytokines. The PIBF gene encodes a hydrophilic protein consisting of 757 amino acids, featuring an N-terminal signal sequence, a leucine zipper motif, a basic zipper sequence, a PEST sequence, a nuclear localization signal, and an endoplasmic reticulum membrane retention signal, along with numerous predicted sites for N-glycosylation and phosphorylation, which are critical for functional regulation and interactions within the cellular environment. Anti-PIBF antibody (C-9) is an invaluable tool for researchers studying the intricate mechanisms of immune modulation during pregnancy and reproductive health implications.
