PDK3 Antibody (RR-2) is a mouse monoclonal IgG2a light chain antibody that detects pyruvate dehydrogenase kinase 3 (PDK3) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). PDK3 plays a crucial role in cellular metabolism by regulating the activity of the pyruvate dehydrogenase complex (PDC), which is essential for converting pyruvate into acetyl-CoA, a key substrate for the citric acid cycle. This regulation is particularly important during metabolic stress, such as starvation, when the body shifts its energy production pathways. PDK3 achieves this by phosphorylating the E1α subunit of PDC, leading to its inactivation, thereby preventing the conversion of pyruvate to acetyl-CoA and promoting fatty acid oxidation instead. The expression of PDK3 is notably high in the testis and is also present in heart and skeletal muscle, indicating its significant role in energy metabolism across different tissues. The protein′s structure includes five conserved regions that are similar to bacterial His-kinases, highlighting its evolutionary significance and the necessity of histidine residues for its kinase activity.
