PCDH15 Antibody (H-3) is a mouse monoclonal IgG3 kappa light chain antibody that detects PCDH15 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-PCDH15 antibody (H-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PCDH15, also known as protocadherin 15, is a critical calcium-dependent cell-adhesion protein that plays a vital role in maintaining normal cochlear and retinal function, making PCDH15 essential for auditory and visual processes. PCDH15 is primarily expressed in the testis, brain, lung, kidney, and spleen, and consists of 1,955 amino acids with a single-pass type I membrane structure featuring 11 cadherin domains. Multiple alternatively spliced isoforms of PCDH15 allow for functional diversity, which is crucial for establishing and maintaining neuronal connections in the brain. Defects in PCDH15 are linked to several conditions, including Usher syndrome type 1F, Usher syndrome type 1D/F, and non-syndromic sensorineural deafness autosomal recessive type 23, highlighting PCDH15′s significance in sensory function.

Is this antibody good for live cell FAC sorting? Or is this antibody target the extra cellular dormain of PCDH15 protein?

Asked by: huangwei0705
Thank you for your question. This antibody has not been tested for cell FAC sorting and can not be recommended or warrantied for that use. The epitope of this antibody maps within amino acids 781-1079 of human PCDH15.

What is the recommended fixation method for immunofluorescence staining with PCDH15 (H-3): sc-377235 monoclonal antibody?

Asked by: jenniferc
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
