PARP-1 Antibody (E-8) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 1-300 at the N-terminus of human PARP-1 and is reactive with mouse, rat, and human samples, making anti-PARP-1 antibody (E-8) suitable for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PARP-1, also called poly(ADP-ribose) polymerase-1, ARTD1, Adprt, and Parp1, is a nuclear DNA-binding protein that plays a critical role in DNA repair, chromatin remodeling, and apoptosis. Located predominantly in the nucleus, PARP-1 is essential for recognizing DNA damage, especially strand breaks, and signaling for repair by catalyzing the transfer of ADP-ribose units from NAD+ to target proteins, including histones and other chromatin-associated proteins. This nuclear localization positions PARP-1 at the heart of genomic maintenance, allowing rapid response to DNA damage and helping prevent mutations that could lead to chromosomal instability. In addition to repair functions, PARP-1 influences transcription and cell death pathways, as cleavage of PARP-1 during apoptosis generates an 89-kDa fragment that translocates to the cytoplasm, contributing to the apoptotic process. PARP-1′s ability to regulate both survival and death pathways underscores PARP-1′s importance in maintaining cellular homeostasis and genome integrity. In conclusion, PARP-1 monoclonal antibody (E-8) enables comprehensive investigation of PARP-1′s essential roles in DNA repair, chromatin remodeling, and apoptosis across multiple species and experimental platforms.

When should the TransCruz reagent sc-74469 X be used instead of the standard form sc-74469?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
