PARP-1 Antibody (B-10) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 1-300 at the N-terminus of human PARP-1 and shows reactivity with mouse, rat, and human samples, making anti-PARP-1 antibody (B-10) suitable for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PARP-1, also called poly(ADP-ribose) polymerase-1, ARTD1, Adprt, and Parp1, is a nuclear DNA-binding protein with a complex structure that includes a DNA-binding domain, an automodification domain, and a catalytic domain. The DNA-binding domain, characterized by zinc finger motifs, enables PARP-1 to detect DNA strand breaks, which is crucial for initiating DNA repair. The automodification domain plays a role in regulating protein-protein interactions, including PARP-1 self-regulation, while the catalytic domain transfers ADP-ribose units from NAD+ to target proteins, such as histones and other chromatin-associated proteins, to facilitate chromatin relaxation and access to the DNA repair machinery. This structured multi-domain organization is essential for PARP-1′s role in maintaining genomic stability, as PARP-1 ensures precise coordination of DNA damage detection, signaling, and repair. The integrity of this structure is critical, as disruptions in PARP-1 function can lead to chromosomal instability, higher rates of chromosome fusions, and aneuploidy, underscoring PARP-1′s importance in safeguarding genome integrity.
