PARD6B encodes a core component of the PAR polarity complex that helps establish and maintain apical–basal polarity in epithelial cells and regulates asymmetric cell division. Through interactions with atypical PKC (aPKC) and other polarity regulators, PARD6B coordinates tight junction assembly, cytoskeletal organization, and directional cell migration, linking polarity control to pathways governing tissue architecture. Disruption of polarity signaling is associated with epithelial disorganization, altered adhesion, and invasive phenotypes, making PARD6B relevant to studies of tumor progression and metastasis. PARD6B-dependent polarity programs are also important during development and differentiation, where spatial control of signaling and junctional integrity shapes organogenesis.
PARD6B CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PARD6B gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PARD6B together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PARD6B open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish PARD6B protein expression.
This CRISPR knockout system enables efficient generation of PARD6B-deficient cell models for investigation of PARD6B signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
