Parafibromin Antibody (2H1) is a mouse monoclonal IgG1 kappa light chain antibody that detects Parafibromin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-Parafibromin antibody (2H1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Parafibromin, encoded by the CDC73 gene, plays a critical role in regulating gene expression and is a key component of the RNA polymerase II/Paf1 complex, which is essential for histone modification and transcriptional regulation. This complex is vital for proper RNA polymerase II functioning, as Parafibromin facilitates histone modification, influencing chromatin structure and gene accessibility. Parafibromin serves as a potential biomarker for diagnosing parathyroid carcinoma, a condition characterized by heterogeneous tumors that can complicate diagnosis. The human CDC73 gene, located on chromosome 1q25, is the homolog of yeast Cdc73 and is implicated in hyperparathyroidism with jaw tumor syndrome (HPT-JT), highlighting Parafibromin′s clinical significance in endocrine tumor suppression.

Detected parafibromin at expected size in 15 ug total proteine lysate, wet transfer one hour at 450 mAmps on PVDF membrane, overnight blocking with 5% nonfat dry milk in PBS, one hour incubation at RT with 1:500 anti Parafibromin in 5% PBS with 0,2%Tween followed by one hour incubation with 1:10.000 anti mouse (sc-358914) HRP conjugated antibody and detection with ECL prime substrate followed by short film exposure (under 1 minute) and automatic processing.
