PANK3 Antibody (MDA-299-62A) is a mouse monoclonal IgG2a antibody that detects PANK3 of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). PANK3 (MDA-299-62A) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The pantothenate kinase (PANK) family, to which PANK3 belongs, is crucial for catalyzing the initial step in the biosynthesis of coenzyme A (CoA), a vital coenzyme that plays a significant role in the synthesis and oxidation of fatty acids, as well as in the oxidation of pyruvate during the citric acid (Krebs) cycle. PANK3, a 370 amino acid protein, is primarily localized in the cytoplasm and is essential for the physiological regulation of intracellular CoA concentrations. PANK3 is subject to feedback inhibition by CoA and its thioesters, which ensures that cellular levels of CoA remain balanced. PANK3 catalyzes the phosphorylation of pantothenate (Vitamin B5) using ATP, resulting in the formation of 4′-phosphopantothenate, a critical intermediate in CoA biosynthesis. Notably, PANK3 is closely related to other family members, including PANK1, PANK2, and PANK4, and is highly expressed in the liver. Mutations in the gene encoding PANK2, the only mitochondria-targeted human PANK, are associated with pantothenate kinase-associated neurodegeneration (PKAN), highlighting the importance of PANK family proteins in human health.
