PANK3, short for Pantothenate Kinase 3, is a protein that in humans is encoded by the PANK3 gene. This protein is a member of the pantothenate kinase family, which are key regulatory enzymes in the biosynthesis of Coenzyme A (CoA), a vital molecule involved in numerous cellular processes, including energy metabolism and the synthesis and degradation of fatty acids.The PANK3 protein catalyzes the first step in CoA biosynthesis, which is the phosphorylation of pantothenate (vitamin B5) to form 4'-phosphopantothenate. This step is crucial in controlling the rate of CoA synthesis.
PANK3 is one of four pantothenate kinases in humans (PANK1, PANK2, PANK3, and PANK4) and is known for its unique tissue-specific expression. While the other PANK enzymes are widely distributed in different tissues, PANK3 expression is primarily restricted to the testes.Despite this specific expression, the exact role and significance of PANK3 in testicular function and overall human physiology are not well understood. The functions of PANK1 and PANK2 have been more extensively studied, and mutations in the PANK2 gene are known to cause a rare neurodegenerative disease called Pantothenate Kinase-Associated Neurodegeneration (PKAN).
