PA28α Antibody (G-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects PA28α in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-PA28α antibody (G-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PA28α functions as an interferon-gamma inducible proteasome activator, essential for presenting specific major histocompatibility class I antigens. PA28α forms a heteropolymer with PA28β, creating a hexameric ring that binds to the ends of the 20S proteasome, enhancing proteolytic activity. This interaction enables efficient peptide processing for MHC class I presentation, which is crucial for immune response. PA28α is constitutively expressed in antigen-presenting cells, and downregulation can lead to abnormal proteasome activation, contributing to conditions such as intimal hyperplasia in animal models. The gene encoding PA28β, known as PSME2, is located on chromosome 14q11.2 and is essential for PA28 complex formation, highlighting the importance of both subunits in maintaining proper immune function.
