p53 Antibody (Pab 240) is an IgG1 kappa mouse monoclonal p53 antibody (also designated p53 antibody or TP53 antibody) that detects only mutant p53 under non-denaturing conditions but is equally reactive with mutant and wildtype p53 under denaturing conditions of mouse, rat and human origin by WB, IP and IF. p53 Antibody (Pab 240) is available as the non-conjugated anti-p53 antibody form, as well as multiple conjugated forms of anti-p53 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. p53, a DNA-binding, oligomerization domain- and transcription activation domain-containing tumor suppressor, upregulates growth arrest and apoptosis-related genes in response to stress signals, thereby influencing programmed cell death, cell differentiation, and cell cycle control mechanisms. p53 localizes to the nucleus, yet can be chaperoned to the cytoplasm by the negative regulator, MDM2. MDM2 is an E3 ubiquitin ligase that is upregulated in the presence of active p53, where it poly-ubiquitinates p53 for proteasome targeting. p53 fluctuates between latent and active DNA-binding conformations and is differentially activated through posttranslational modifications, including phosphorylation and acetylation. Mutations in the DNA-binding domain (DBD) of p53, amino acids 110-286, can compromise energetically-favorable association with cis elements and are implicated in several human cancers.

I am trying to do immunoprecipitation using this antibody. but I am not getting any pull however the p53 Do1 antibody gives a good band. I use 1ug of antibody and I am using p53 recombinant protein 1ug. I have tried PBS and NP-40 buffer with 1% NP-40.

I am looking to perform immunofluorescence studies in mice tissue samples. Is the p53 Antibody (Pab 240): sc-99 a viable option for this?

If looking to perform immunofluorescence in mice tissue samples, we recommend using our directly conjugated mouse monoclonal primary antibodies. The p53 Antibody (Pab 240): sc-99 is NOT currently available with direct conjugation, but we do offer an alternative antibody, p53 Antibody (DO-1): sc-126. This alternative (sc-126) is currently available conjugated to HRP (sc-126 HRP), phycoerythrin (sc-126 PE), fluorescein (sc-126 FITC), Alexa Fluor® 488 (sc-126 AF488), Alexa Fluor® 647 (sc-126 AF647), biotin (sc-126 B), TRITC (sc-126 TRITC, 200 µg/ml), PerCP (sc-126 PerCP), PerCP-Cy5.5 (sc-126 PCPC5), or Alexa Fluor® 405 (sc-126 AF405).

Rated 4 out of 5 by from A little faint, but does workI used this with human lysates for western blot. Band was a little faint, but it definitely does work. I've heard lysate prep for p53 can be tricky, so I may need to just improve my lysis buffer recipe/protocol.

Rated 5 out of 5 by from Great for Western blot!We used this antibody at a 1:1000 dilution to detect the DNA-binding domain of recombinant p53 from E.coli expression. The antibody works great and there are clear, distinct bands on the gel. Would definitely recommend!

Rated 5 out of 5 by from strong band at 53kdaThis antibody been used to detect the p53 protein in mouse lung tissue.I have optimizing antibody concentrations from 1:300 to 1:1000,I prefer to use 1:500 ,this dilution got clean band at 53kda and no background.If the background is too high, I recommend use 5% BSA for blocking to reducing background.
