p53 Antibody (Pab 1801) is an IgG1 kappa mouse monoclonal p53 antibody (also designated TP53 antibody) that detects p53 of mouse, rat and human origin by WB, IP, IF, IHC(P) and FCM. p53 Antibody (Pab 1801) is available as both conjugated and non-conjugated anti-p53 antibody forms. p53, a DNA-binding, oligomerization domain- and transcription activation domain-containing tumor suppressor, upregulates growth arrest and apoptosis-related genes in response to stress signals, thereby influencing programmed cell death, cell differentiation, and cell cycle control mechanisms. p53 localizes to the nucleus, yet can be chaperoned to the cytoplasm by the negative regulator, MDM2. MDM2 is an E3 ubiquitin ligase that is upregulated in the presence of active p53, where it poly-ubiquitinates p53 for proteasome targeting. p53 fluctuates between latent and active DNA-binding conformations and is differentially activated through posttranslational modifications, including phosphorylation and acetylation. Mutations in the DNA-binding domain (DBD) of p53, amino acids 110-286, can compromise energetically-favorable association with cis elements and are implicated in several human cancers.

Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue?

This antibody is currently available conjugated to phycoerythrin (sc-98 PE), Alexa Fluor® 488 (sc-98 AF488) or Alexa Fluor® 647 (sc-98 AF647). Rated 1 out of 5 by from IHC on mouse tissue not workingI tried this antibody on immunoperoxidase staining of formalin-fixed, paraffin-embedded mouse breast carcinoma tissue at 1/50 and it did not work. No staining.

This gave a single, clear band, although I had to use a higher titer of antibody (1:100 dilution) to detect.
