p53 Antibody (D-11) is a mouse monoclonal antibody of the IgG1 kappa chain, raised against the full-length human p53 protein (amino acids 1–393), and specifically detects p53 from human, mouse, and rat origins in applications such as western blotting, immunoprecipitation, immunofluorescence, and enzyme-linked immunosorbent assay. p53 mouse monoclonal antibody (D-11) is widely utilized in research focused on the tumor suppressor p53, an important DNA-binding protein with oligomerization and transcription activation domains, which upregulates genes associated with cell cycle arrest and apoptosis in response to cellular stress, thereby playing a critical role in programmed cell death, cell differentiation, and cell cycle control mechanisms; p53 is primarily localized in the nucleus but can be exported to the cytoplasm by its negative regulator MDM2, an E3 ubiquitin ligase that promotes p53 degradation via the proteasome pathway. Under normal conditions, p53 levels within the cell are kept low through continuous degradation mediated by the MDM2 protein, an E3 ubiquitin ligase that targets p53 for proteasomal degradation. However, when cells experience stress signals such as DNA damage, oncogene activation, hypoxia, or nucleotide depletion, p53 becomes stabilized and accumulates in the nucleus. There, p53 functions as a transcription factor, activating or repressing a wide array of genes involved in cell cycle arrest, DNA repair, senescence, and programmed cell death (apoptosis). Further, the functional activity of p53 is modulated by post-translational modifications such as phosphorylation and acetylation, and mutations within its DNA-binding domain (amino acids 110–286) can impair its DNA-binding capacity, leading to compromised tumor suppressor functions and are implicated in numerous human cancers.

what is the difference between C-11 and D-11?

Thank you for your question. C-11 and D-11 are monoclonal IgG1 antibodies produced by two different clones. The antibodies have both been raised against amino acids 1-393 representing full length p53 of human origin, however they bind against different specific epitopes of the same target protein and may exhibit different binding characteristics.

What is the recommended fixation method for immunofluorescence staining with p53 (D-11): sc-17846 monoclonal antibody?
