p53 Antibody (C-11) is an IgG1 kappa light chain mouse monoclonal antibody designed to detect p53 from human, mouse, and rat origins in applications such as western blotting, immunoprecipitation, immunofluorescence, and enzyme-linked immunosorbent assay. p53 monoclonal antibody (C-11) is highly specific to p53, raised against the full-length of human p53 protein, and is available in multiple forms, including non-conjugated and various conjugated versions like agarose, horseradish peroxidase, fluorescein isothiocyanate, phycoerythrin, and multiple Alexa Fluor® conjugates. The p53 protein is a crucial tumor suppressor protein containing DNA-binding, oligomerization, and transcription activation domains. P53 upregulates genes related to growth arrest and apoptosis in response to stress signals, thereby influencing programmed cell death, cell differentiation, and cell cycle control mechanisms. Typically localized in the nucleus, p53 can be transported to the cytoplasm by its negative regulator MDM2, an E3 ubiquitin ligase that is upregulated in the presence of active p53 and targets it for proteasomal degradation through poly-ubiquitination. The activity of p53 oscillates between latent and active DNA-binding conformations and is modulated by post-translational modifications such as phosphorylation and acetylation. Mutations within the DNA-binding domain of p53 (amino acids 110–286) can impair its DNA-binding capability and are implicated in various human cancers.
