P2Y5 CRISPR/Cas9 KO Plasmid (h) | sc-405189 | 20 µg | $397.00 |
LPAR6 (also known as P2Y5) encodes a G protein–coupled receptor that responds to lysophosphatidic acid to regulate intracellular signaling through heterotrimeric G proteins. P2Y5 activity influences Rho/ROCK- and MAPK-linked pathways, coordinating cytoskeletal remodeling, cell–cell adhesion, and motility in a context-dependent manner. The receptor is prominently implicated in skin and hair follicle biology, where altered LPAR6 signaling is associated with congenital hair growth disorders and broader epithelial homeostasis phenotypes. In addition, LPAR6-mediated lipid signaling intersects with inflammatory and barrier-related processes, making it relevant for mechanistic studies in keratinocytes and other epithelial models.
P2Y5 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the LPAR6 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the LPAR6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the LPAR6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish P2Y5 protein expression.
This CRISPR knockout system enables efficient generation of LPAR6-deficient cell models for investigation of P2Y5 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
