p16 INK4A Antibody (JC8) is a mouse monoclonal IgG2a antibody that detects p16 INK4A of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). p16 INK4A (JC8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. p16 INK4A functions as a tumor suppressor by inhibiting cyclin-dependent kinase 4 (Cdk4), thereby preventing retinoblastoma protein (Rb) phosphorylation and blocking cell cycle progression from G1 phase to S phase. This regulatory function maintains proper cell cycle control and prevents uncontrolled cell proliferation, a hallmark of cancer. p16 INK4A gene alterations, including homozygous deletions and point mutations, are frequently found in various human tumors, highlighting its significance in tumorigenesis. Using anti-p16 INK4A antibody (JC8) enables researchers to investigate cell cycle regulation mechanisms and p16 INK4A dysregulation in cancer biology.

Dopo l'utilizzo di questo anticorpo per un western blot è stato possibile osservare la comparsa di due bande in ogni replicato che ho analizzato. Vorrei sapere a cosa corrispondono le due differenti bande

Does this antibody apply to mouse or rat tissue for WB or IHC?

Rated 1 out of 5 by from Non funziona su estratti proteici di topoHo provato la trial size circa un mese fa. Purtroppo non funziona per western blot su estratti proteici preparati da muscolo di topo. Non riconosce la proteina P16 nè nel muscolo di topo WT, nè in di topo trangenico che dovrebbero avere un'induzione di P16. Riconosce solo le catene leggere e pesanti delle immunoglobuline a 20 kDa e 55kDa, rispettivamente

Rated 5 out of 5 by from Western blot excellent detectionHeLa lysate, 40ug total protein load per well, semi-dry transfer, 30 minute block, primary overnight 4 degrees C, secondary goat anti-mouse IgG (H+L) cross-adsorbed HRP 1:10,000 1hr RT, ECL substrate, GeneGnome gel imager photograph. Block and antibody solutions: 5% blotting-grade block in TBST. Excellent detection. May try lower concentration.

Rated 5 out of 5 by from very good signal by WBI used this antibody routinely for the detection of p16 in primary keratinocytes during replicative senescence (1:1000 in 5% milk/PBST ON 4C). MW around 15-20 kDa (using precast or >12% gels). We used primary keratinocytes at passage 4 as positive control of the antibody.
