SEE ALSO...

p-p53 Antibody (B-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Thr 55 phosphorylated p53 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). p-p53 monoclonal antibody (B-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. p53 plays a crucial role as a tumor suppressor, primarily functioning to regulate cell cycle and promote apoptosis in response to cellular stress, thereby preventing proliferation of potentially cancerous cells. Nuclear localization of p53 is vital for function, as p53 binds to DNA to activate genes involved in growth arrest and apoptosis. MDM2, an E3 ubiquitin ligase, can sequester p53 in the cytoplasm for degradation, highlighting the importance of nuclear presence for tumor suppression. Tetramer formation and dynamic conformational changes of p53, influenced by post-translational modifications such as phosphorylation and acetylation, are critical for activity. Mutations in the DNA-binding domain of p53 can disrupt function and are frequently associated with various human cancers, underscoring the significance of monitoring p53 activity and modifications in cancer research. These attributes make anti-p53 antibody (B-3) a critical tool for studying regulation, activity, and modifications of p53 in both normal cellular processes and cancer pathogenesis.

Hello, We are conducting a western blot using fibroblast cells and we are using the p-p53 antibodies (B-3): sc-377553 to detect phosphorylated p53 (Thr 55). We were wondering what size the bands show up as since we don't really detect 50 kDa bands.

Asked by: Anonymous
Thank you for your question. The expected molecular weight of p-p53 is 53 kDa. If issues in your experiment persist, please reach out to our technical service team for direct troubleshooting assistance.

This antibody (sc-377553) seems to recognize a higher band than predicted (around 90 kDa vs 55 kDa). I was wondering if you could comment on that. Other antibodies against P-p53 seem to detect a protein around 55 kDa.

What application is the blocking peptide sc-377553 P appropriate for?

Asked by: TinTin
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

Rated 3 out of 5 by from 抗体的非特异信号多我们使用这个抗体做western 实验,实验结果显示背景特别高,主带信号弱,杂带很多。尽管我们使用了BSA 作为封闭剂,且封闭2个小时,但是杂带还是很多。

Rated 3 out of 5 by from an OK antibody for WBThis antibody works for WB (1:500 dilution, 40 ug HeLa Lysate), although the signal intensity is low (and also with a higher background). However, we used frozen cell lysates and have not tested fresh samples.
