p-Lck Antibody (pY505.4) is a mouse monoclonal IgG1 antibody that detects Lck in mouse, rat, and human samples through western blotting (WB) and immunoprecipitation (IP) applications. p-Lck (pY505.4) antibody specifically recognizes the phosphorylated form of Lck at tyrosine 505, a critical regulatory site that modulates the kinase′s activity. Lck, a member of the Src family of protein tyrosine kinases, plays a pivotal role in T cell signaling and activation, making Lck essential for immune responses. The phosphorylation of Lck at Tyr 505 by Csk serves as a negative regulatory mechanism, inhibiting catalytic activity under certain conditions, while phosphorylation at Tyr 394 enhances activity, promoting T cell activation. This dynamic regulation of Lck is crucial for maintaining the balance between T cell activation and inhibition, which is vital for preventing autoimmune diseases and ensuring effective immune responses. Additionally, Lck is predominantly localized in the plasma membrane of T cells, where Lck interacts with various signaling molecules, including the T cell receptor, to initiate downstream signaling cascades. I am using p-Lck (pY505.4): sc-136184 antibody for immunoprecipitation, how much antibody should I use?

Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
