Otulin encodes OTULIN, a deubiquitinase that selectively hydrolyzes Met1 (linear)-linked ubiquitin chains generated by the LUBAC complex, thereby shaping ubiquitin-dependent signal propagation. In mouse cells, OTULIN functions as a key negative regulator of NF-κB and inflammatory signaling by controlling linear ubiquitination of pathway components such as RIPK1 and NEMO/IKBKG, influencing cytokine responses and cell death decisions. Disruption of OTULIN perturbs proteostasis and immune homeostasis, linking this pathway to autoinflammatory phenotypes and broader inflammatory disease mechanisms. OTULIN also intersects with innate immune signaling and stress responses where linear ubiquitin editing tunes the amplitude and duration of receptor-triggered signaling.
OTULIN CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Otulin gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Otulin together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Otulin open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish OTULIN protein expression.
