OSX Antibody (F-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects OSX in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-OSX antibody (F-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. OSX, also known as Osterix, is a crucial zinc finger-containing transcriptional activator that plays a vital role in osteoblast differentiation, which is essential for bone formation. OSX is expressed in all developing bones and regulates the expression of genes necessary for osteoblast maturation. Nuclear localization allows OSX to bind to GC box promoter elements, thereby activating mRNA synthesis from target genes. Absence of OSX leads to a failure in the differentiation of periosteal and mesenchymal cells into osteoblasts, resulting in significant impairment in bone matrix deposition and bone formation. Studies have shown that deficiency of cyclooxygenase-2 (Cox-2) correlates with reduced OSX expression, indicating that Cox-2 may play a role in inducing OSX to facilitate skeletal repair. This relationship highlights OSX′s importance not only in bone development but also in potential recovery processes following skeletal injuries.

Which secondary antibodies do you recommend for IF staining in mouse tissues?

You can find our anti-mouse secondary binding proteins by following this link: https://www.scbt.com/browse/Secondary-Antibodies/_/N-1hoaxk4 OSX Antibody (F-3): sc-393325 is a kappa light chain antibody, so any of our kapppa binding proteins such as m-IgGκ BP-CFL 488 sc-516176 would be suitable.

Are there any issues with non-specific staining when using this mouse monoclonal antibody on mouse tissue samples in IF or IHC?

Thank you for your question. This antibody is recommended for use with mouse samples in IF and IHC. To eliminate any potential issues with non-specific binding of the anti-mouse secondary, we recommend using the directly conjugated version of the primary. This antibody is available conjugated to PE, FITC, AF488 and AF647.
