O-GlcNAc transferase Antibody (F-12) is a mouse monoclonal IgG2a kappa light chain antibody that detects O-GlcNAc transferase of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). O-GlcNAc transferase (F-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. O-linked N-acetylglucosamine transferase, also known as OGT, plays a crucial role in cellular signaling by catalyzing the addition of a single N-acetylglucosamine moiety to serine or threonine residues on target proteins. This post-translational modification influences protein function, stability, and interactions, thereby impacting various cellular processes such as transcription, cell cycle regulation, and stress responses. The dynamic interplay between O-GlcNAcylation and phosphorylation at these residues is particularly important, as they can compete for the same sites, leading to functional outcomes that are critical for maintaining cellular homeostasis. O-GlcNAc transferase is expressed in multiple human tissues, with the pancreas showing the highest levels, and exists as a heterotrimeric complex, indicating potential involvement in complex regulatory networks within the cell. Immunofluorescence (IF) studies have shown that O-GlcNAc transferase localizes in both the nucleus and cytosol, suggesting a multifaceted role in cellular functions. Notably, prolonged expression of O-GlcNAc transferase in HeLa cells has been associated with reduced cell viability, highlighting potential toxicity and the need for careful consideration in experimental designs.

Can you recommend a positive control to use with O-GlcNAc transferase (F-12): sc-74546 monoclonal antibody for Western blot?

We recommend SK-N-MC nuclear extract (sc-2154), A549 (sc-2413) or c4 (sc-364186) whole cell lysates to use as positive controls as we have Western blot data showing specific detection of O-GlcNAc transferase in these cell lines.

Rated 4 out of 5 by from Good signal in ImmunofluorescenceWe used this Antibody in Immunosfluorescence (IF) of single mouse zygotes at a 1:200 dilution and we detected good signal compared to a secondary antibody only control. Specificilty cannot be evaluated by this experiment because IF does not give information about size and we did not have an Ogt-KO control sample.
