Neuropeptide Y (NPY) encodes a 36–amino acid secreted neuropeptide that signals through G protein–coupled Y receptors to regulate neurotransmission, neuroendocrine secretion, and autonomic control of energy balance. NPY activity couples to Gi/o-mediated signaling to modulate cAMP levels, calcium flux, and downstream kinase pathways that influence neuronal excitability, synaptic plasticity, and appetite-related circuitry. Beyond the central nervous system, NPY contributes to stress responses, vascular tone, and immune cell behavior, linking it to processes such as inflammation and metabolic homeostasis. Dysregulated NPY signaling has been studied in contexts including obesity and metabolic disease, anxiety- and stress-related phenotypes, and neurodegenerative or neuroinflammatory conditions.
NPY CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the NPY gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the NPY together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the NPY open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish NPY protein expression.
This CRISPR knockout system enables efficient generation of NPY-deficient cell models for investigation of NPY signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
