SEE ALSO...

Nogo Antibody (C-4) is a mouse monoclonal IgM antibody that detects Nogo protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Nogo antibody (C-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Nogo protein, also known as neurite outgrowth inhibitor or neuroendocrine secretion regulating protein, plays a critical role in the central nervous system by inhibiting axonal growth, which is essential for maintaining the integrity of CNS white matter. Nogo is primarily expressed by oligodendrocytes and is associated with the endoplasmic reticulum, where Nogo exists in three splice variants: Nogo-A, Nogo-B, and Nogo-C. Nogo presence in the CNS is crucial as Nogo prevents excessive axonal regeneration, which could lead to aberrant neural connections and potential dysfunction. Understanding Nogo interactions with other proteins, such as the NgR (Nogo receptor), is vital for elucidating Nogo′s role in neuroregeneration and the development of therapeutic strategies for spinal cord injuries and neurodegenerative diseases.

Can Nogo Antibody (C-4), sc-271878 be used for immunoprecipitation? If so can a Protein A/G beads be used to pull down?

Asked by: Atish
Thank you for your question. Yes, Nogo (C-4) can be used for IP and we recommend using protien L Agarose. https://www.scbt.com/p/protein-l-agarose

What region of Nogo-A, B, and C does this antibody recognize? I have a KO mouse with a flxed region of Exon 4/5 ( first two common exons between all Nogo proteins). I am still seeing Nogo expression in cells where Nogo should be KO. Confirmed KO via PCR

The sample blot shows a protein of about 55 kDa, corresponding to the Nogo B isoform, but no band around 125 kDa, the molecular weight of the Nogo A isoform. Is this antibody preference or differential expression of the isoforms in HEK cells?

Asked by: shoran
Thank you for your question. The Western blot data shown on our website is representative of what you can expect when using this antibody. We haven't tested for differential isoform expression in HEK cells. There is Western blot data in Neuro-2A and EOC20 whole cell lysates showing a signal detected around 220 kDa, the expected observed molecular weight for Nogo A.

What is the Molecular Weight of the Nogo Antibody (C-4)?

Asked by: asj64
We do not determine the exact molecular weight for each individual antibody. However, the expected molecular weight detected by this Nogo Antibody (C-4): sc-271878 can be found on our product datasheet. A link is provided below. https://datasheets.scbt.com/sc-271878.pdf

What application is the blocking peptide sc-271878 P appropriate for?

Asked by: Randy McDonald
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

Rated 5 out of 5 by from Great Antibody!I tested 30-32 antibodies from Santa Cruz. I used the Nogo Antibody (C-4) to detect RTN4 from mouse brain lysates. I detected a single band in WB (1:200).

Rated 5 out of 5 by from Excellent for NogoA & B, but not for NoGo CIt's an excellent antibody for detection of NogoA and NogoB in western blot and IF. I used 1:60 dilution of this antibody in IF, for staining Nogo A & B in PFA fixed HeLa cells. I used 1:200 dilution in western blot for detecting NogoA & B in HeLa cell lysate. There was a strong band for NogoB and a faint band for NogoA, but no band for NogoC.

Rated 5 out of 5 by from Great antibodyWorked well for IF staining of retinal cross sections from mouse.
Date published: 2017-09-18
Rated 3 out of 5 by from Not usable on J774A.1 cellsProduces only very dim diffuse staining of J774A.1 cells. On the other hand, it was a free sample, so no harm done.
