Nmi Antibody (XX-22) is a mouse monoclonal IgG2b antibody that detects Nmi in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Nmi, or N-Myc interactor, plays a crucial role in cellular signaling and transcriptional regulation, particularly in myeloid leukemias and other cancers where Nmi is often overexpressed. Nmi is primarily located in the nucleus, where Nmi interacts with several key transcription factors, including c-Myc, n-Myc, and members of the Stat family, thereby influencing gene expression in response to cytokine stimulation. Nuclear localization of Nmi is vital as Nmi allows for direct modulation of transcriptional activity, enhancing the function of Stat proteins by recruiting essential coactivators such as CREB-binding protein (CBP) and p300. This recruitment significantly increases transcriptional responsiveness of Stat proteins, particularly Stat5, to interleukin-2 stimulation, underscoring Nmi′s importance in regulating immune responses and cellular growth. With an ability to enhance transcriptional activity despite lacking intrinsic DNA binding capabilities, Nmi serves as a critical mediator in signaling pathways that govern cell proliferation and differentiation, making Nmi a valuable target for research in cancer biology and therapeutic development.

Thank you for your question. Yes, sc-101100: Nmi (XX-22) is recommended for use in IHC with paraffin-embedded sections.
