Nmi Antibody (D-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects Nmi protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Nmi (D-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Nmi protein, known as N-Myc interactor, plays a crucial role in cellular signaling and transcriptional regulation, particularly in myeloid leukemias and c-Myc overexpression. Nmi functions as an interferon-inducible protein that interacts with several transcription factors, including c-Myc, n-Myc, Max, and c-Fos, which possess bHLH-ZIP, bHLH, or Zip domains. Nmi expression remains low across various fetal and adult tissues but shows significant upregulation in myeloid leukemias. Nmi enhances transcriptional activity of the Stat family of transcription factors, such as Stat1 and Stat5, by recruiting coactivators like CREB-binding protein (CBP) and p300, thereby amplifying Stat-dependent transcription in response to cytokine stimulation. This interaction regulates immune responses and cellular growth by increasing Stat proteins′ affinity for coactivators, leading to a five-fold enhancement in transcriptional responsiveness to interleukin-2 stimulation when Nmi co-expresses with CBP.
