Cd244 encodes NK 2B4 (CD244), a SLAM family immunoreceptor expressed on mouse natural killer cells and subsets of T cells that modulates cytotoxicity and cytokine production through interactions with CD48. Signaling is mediated by cytoplasmic ITSM motifs that recruit adaptor proteins such as SAP/SH2D1A or EAT-2, shaping downstream phosphorylation networks that tune activating versus inhibitory outputs. Through these pathways, CD244 contributes to immune synapse formation, target cell recognition, and regulation of inflammatory responses. Altered CD244 signaling has been linked to dysregulated antiviral and antitumor immunity, NK cell functional exhaustion, and immune imbalance in settings of chronic inflammation and hematologic malignancy models.
NK 2B4 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Cd244 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Cd244 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Cd244 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish NK 2B4 protein expression.
This CRISPR knockout system enables efficient generation of Cd244-deficient cell models for investigation of NK 2B4 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
