Ninein Antibody (F-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects Ninein protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Ninein antibody (F-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Ninein plays a crucial role in organizing and stabilizing the microtubule network, particularly at the centrosome, where Ninein is essential for proper positioning and anchorage of the microtubule minus-end in epithelial cells. This function is vital for maintaining cellular architecture and facilitating processes such as cell division and intracellular transport. Ninein acts as a centrosome maturation factor and participates in microtubule nucleation, although Ninein overexpression doesn′t enhance nucleation or elongation; instead, suppressing microtubule release. Ninein interacts with glycogen synthase kinase 3 beta (GSK3B) through its C-terminal domain and associates with C14orf166, which inhibits Ninein phosphorylation by GSK-3β. Structurally, Ninein organizes in a tubular conformation within the core centrosome, with open and closed ends contained inside, and shows ubiquitous expression, with predominant levels in heart and skeletal muscle tissues. The coiled-coil region spanning from Asn 1611 to Pro 1693 is critical for targeting Ninein to the centrosome, underscoring Ninein′s importance in cellular function and organization.
