Neutral ceramidase Antibody (B-9) is a mouse monoclonal IgG2b kappa light chain antibody that detects neutral ceramidase of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-neutral ceramidase antibody (B-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Neutral ceramidase, also known as ASAH2 (N-acylsphingosine amidohydrolase (non-lysosomal ceramidase) 2), HNAC1, BCDase, or NCDase, is a 780 amino acid single-pass type II membrane protein that undergoes proteolytic cleavage to yield a soluble active peptide. Neutral ceramidase is predominantly expressed in the heart, intestine, kidney, and skeletal muscle, where neutral ceramidase plays a crucial role in catalyzing the hydrolysis of sphingolipid ceramide into sphingosine and free fatty acids, a reaction vital for regulating sphingolipid signaling metabolites. The optimal pH for this enzymatic activity ranges from 6.5 to 8.5, highlighting neutral ceramidase′s importance in maintaining cellular homeostasis. Neutral ceramidase is implicated in apoptotic suppression and the digestion of dietary sphingolipids within intestinal tissues. Multiple isoforms of neutral ceramidase exist due to alternative splicing, which may contribute to diverse functional roles in various biological processes.
