Nestin Antibody (10c2) is a mouse monoclonal IgG1 kappa light chain antibody that detects nestin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Anti-nestin antibody (10c2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Nestin is a crucial intermediate filament protein predominantly found in embryonic central nervous system progenitor cells, playing a vital role in structural integrity and function. Nestin expression is essential during muscle development, where nestin interacts with other intermediate filament proteins such as Desmin and Vimentin, forming a dynamic network that supports cellular architecture. Nestin′s ability to co-assemble with Vimentin or α-internexin into heterodimer coiled-coil structures leads to the formation of 10 nm intermediate filaments critical for maintaining cellular shape and resilience. Deletion of nestin in the intermediate filament consensus rod domain can disrupt nestin localization in central nervous system precursor cells and radial glial cells, which may affect neurodevelopmental processes. Nestin serves as a marker for neuroepithelial stem cells, glioma cells, and tumor endothelial cells during periods of rapid growth, highlighting nestin′s importance in both normal development and pathological conditions. During neuronal differentiation, nestin localizes to growth cones, suggesting a role in guiding axon elongation, which is crucial for proper neural network formation. Nestin expression is observed in the zona fasciculata and zona reticularis of rat adrenal gland, as well as in dermatomal cells and myoblasts during early stages of myogenesis, further underscoring nestin′s diverse roles in various tissues.
