STK38 encodes the serine/threonine kinase NDR1, an AGC family kinase that functions downstream of MST1/2 in Hippo-related signaling to regulate cell-cycle progression, centrosome biology, apoptosis, and cytoskeletal organization. NDR1 activity is controlled by phosphorylation and cofactor binding (including MOB proteins), linking upstream stress and polarity cues to transcriptional and structural outputs. Through roles in mitotic control, DNA damage–associated responses, and epithelial homeostasis, STK38/NDR1 has been studied in contexts relevant to tumor biology, neurobiology, and inflammation-related signaling. Perturbation of NDR1-dependent pathways can influence proliferation, survival, and migration phenotypes that are commonly interrogated in mechanistic disease models.
NDR1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the STK38 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the STK38 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the STK38 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish NDR1 protein expression.
This CRISPR knockout system enables efficient generation of STK38-deficient cell models for investigation of NDR1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
